1. Overview
Click Chemistry describes rapid, selective "click" reactions between paired functional groups in mild aqueous solutions. This concept has evolved into a convenient, versatile, and reliable two-step coupling procedure for molecules A and B, widely adopted in biosciences, drug discovery, and materials science.
PuriMag™ G-Alkyne Alkyne-Functionalized Magnetic Nanoparticles are uniform, polymer-coated superparamagnetic nanoparticles. Their hydrophilic surface ensures excellent dispersibility, low non-specific adsorption, and easy handling in diverse buffers. The beads feature a high-density surface coating of alkyne functional groups, enabling covalent capture of functionalized proteins via Cu(I)-catalyzed Azide-Alkyne Cycloaddition (CuAAC).
Workflow Requirements:
Target proteins must be metabolically, enzymatically, or chemically tagged with azide groups.
Post-coupling rigorous washing eliminates non-specifically bound proteins.
Unique surface chemistry ensures ultra-low background binding without surfactants.

3. Instructions for Use
1.Add ~10 mg PuriMag™ Alkyne Magnetic Nanoparticles to 25 mL deionized water.
2.After brief ultrasonication, add:
5 mg copper sulfate solution (5 mg in 0.5 mL H₂O)
22 mg sodium ascorbate solution (22 mg in 1 mL H₂O)
Stir mixture for 30 min.
3.Add azide-functionalized compound, stir overnight at RT.
4.Recover product via magnetic separation. Wash with:
Distilled water
PBS (pH 7.0)
5.Treat final product with EDTA to remove copper ions.
